PrimerBac is a searchable database of taxon-specific PCR primers for microbiome identification. The current release compiles validated primer sets for gut-associated taxa, while the underlying workflow is modular and can be extended to other microbial ecosystems by replacing the reference genome and validation datasets.
Firstly, genome assemblies are retrieved from NCBI based on the target taxon. Pan-genome analysis is then performed to identify conserved, classification-informative genes, followed by primer design and validation against a curated reference dataset. The same workflow can be adapted to gut, oral, soil, aquatic, or other microbiome datasets.
