How to use PrimerBac
PrimerBac is a searchable resource of taxon-specific PCR primer candidates for microbiome identification. It provides species- and genus-level primer sequences, design metrics, and a curated gut-associated reference background. Candidate primers were designed from representative sequences of core orthologous groups present in more than 95% of the included RefSeq genomes and screened against RI before database inclusion. Representative species-level pairs were additionally assessed using broad and temporally independent RefSeq assembly panels.

PrimerBac construction workflow
RefSeq genomes matched to RI taxa were used for pan-genome analysis. Representative sequences from core orthologous groups present in more than 95% of the included genomes were supplied to Primer3 for candidate primer design. Candidate pairs were then screened against the RI background before database inclusion. The modular workflow can be adapted to other microbiome-associated taxonomic collections.
